During the manufacturing process of mAbs, they are subjected to multiple post-translational modifications (PTMs) such as deamidation, oxidation, isomerization, and clipping. The multi-attribute method (MAM) based on LC-MS has emerged as a powerful technique to directly monitor multiple product quality attributes (PQAs) of mAbs simultaneously, while also aligning with “quality by design” (QbD) principles. One of the challenges with the implementation of MAM relates to the variability of sample preparation methods such as protein denaturation, reduction, alkylation, tryptic digestion, and SPE-based sample cleanup. To address this variability, sample preparation has been performed using the Andrew+ Robot, leading to reduced preparation-to-preparation variability.
